چکیده

The method using lucigenin-dependent phagocytic chemiluminescence is described for the assessment of alveolar macrophage metabolic activity in response to stimulation by opsonised particles or soluble agents. The requirement for superoxide anion (O2-) in the production of chemiluminescence is suggested by inhibition (95%) using superoxide dismutase. The results obtained are correlated with those obtained using another method of detecting O2- release (r = 0.61; p less than 0.05) and are also related by regression analysis to polymorphonuclear leucocyte contamination of the alveolar macrophage suspension. This shows that alveolar macrophages produce lucigenin-dependent chemiluminescence of the same order of magnitude as do polymorphonuclear leucocytes.

Human bronchoalveolar lavage cells and luminol-dependent chemiluminescence.

Human bronchoalveolar lavage cells from several different disease states were examined by the technique of zymosan-stimulated, luminol-dependent chemiluminescence. Light production correlated well with polymorphonuclear leucocyte contamination of the alveolar macrophage suspension but not with lymphocyte contamination. Regression analysis indicated that human alveolar macrophages produce little...

Oxidative response of human neutrophils, monocytes, and alveolar macrophages induced by unopsonized surface-adherent Staphylococcus aureus.

In contrast to results with bacterial suspensions, phagocytosis of unopsonized bacteria readily occurs when bacteria are adhered to glass or plastic surfaces. However, in contrast to neutrophils, alveolar macrophages produced much less DNA denaturation as measured by acridine orange metachromasia of phagocytized Staphylococcus aureus. We have studied the phagocytosis of unopsonized surface-adhe...

Assessment of pulmonary macrophage and neutrophil function in sequential bronchoalveolar lavage aspirates in sarcoidosis.

Bronchoalveolar lavage was performed in eight patients with biopsy proved sarcoidosis and eight control subjects matched for age and smoking habit, three sequential 60 ml aliquots of sterile saline being used. Each aliquot was aspirated and analysed separately to determine total and differential cell counts. Cellular metabolic activity was stimulated with latex and measured by means of luminol ...

Rat surfactant protein D enhances the production of oxygen radicals by rat alveolar macrophages.

Rat surfactant protein D (SP-D) was shown to enhance the production of oxygen radicals by rat alveolar macrophages. This enhancement, which was determined by a lucigenin-dependent chemiluminescence assay, was maximal after 18 min at an SP-D concentration of 0.2 micrograms/ml. Surfactant lipids did not influence the stimulation of alveolar macrophages by SP-D, whereas the oxygen-radical producti...

The reversible effect of lignocaine on the stimulated metabolic activity of bronchoalveolar lavage cells.

Lignocaine concentrations were measured in the aspirate from a low volume (100 ml) bronchoalveolar lavage (BAL) in twenty patients who had received topical 4% lignocaine as required, and compared to those found in the aspirate from a 180 ml BAL in ten patients who had received 1.5% isotonic lignocaine. The median BAL supernatant lignocaine concentration was significantly lower at 0.14 mM (range...
